集成应用中试规模的膜分离耦联柱层析工艺进行花色苷提取物的纯化、富集,研究各工艺对花色苷的纯化富集效果以及品质的影响,建立超滤(ultrafiltration,UF)-反渗透(reverse osmosis,RO)-XAD-7HP型大孔树脂耦联的纯化工艺.结果显示:在澄清工艺中,无机陶瓷膜效果较好,最佳工艺为陶瓷膜跨膜压力为0.3 MPa、循环流量为50 L/min;在浓缩工艺中,反渗透膜最佳参数为跨模压力为1.1 MPa、循环流量为23.3 L/min;利用XAD-7HP进行富集.处理后所获得的花色苷提取物纯度大于60%,并且此工艺对花色苷组成、水溶液色泽、pH均没有显著影响.

Abstract:

An integrated membrane process involving ultrafiltration (UF) and reverse osmosis (RO) was developed for clarification and concentration of the anthocyanins extract, and the influence of every process was investigated. The ceramic UF membrane showed better results than the polyether sulfone membrane in clarification. The optimal conditions were trans-membrane pressure 0.3 MPa, axial feed flow rate 50 L/min using ceramic UF membrane module, and the optimal conditions were trans-membrane pressure 1.1 MPa, axial feed flow rate 23.3 L/min with RO membrane in concentration. Then, XAD-7HP resin was used for anthocyanins further purification. The results show that the purity of the anthocyanins is more than 60% after processing and the processes have a non-significant impact on the composition of anthocyanins, color characteristics and pH.
