反应条件对DBBF修饰猪血红蛋白性能的影响

反应条件对DBBF修饰猪血红蛋白性能的影响

猪血红蛋白在脱氧条件下经双(3,5-二溴水杨酸)延胡索酸酯(DBBF)修饰,其携氧能力得到改善,氧合曲线较未经修饰血红蛋白右移,半饱和氧分压(P50值)为3.1 kPa,希尔系数值为2.2,两者都接近人全血值;修饰过程中别构效应调节剂对修饰产物携氧性能影响很大,在反应混合物中加入5.0 mmol/L六磷酸肌醇,产物的P50明显增加.DBBF与血红蛋白的量比为2∶1,反应时间2 h时,产物的P50和希尔系数均接近人体生理指标.电泳图谱分析表明,无基质猪血红蛋白可完全解聚为单体,只在相对分子质量为15×103处出现条带,而DBBF修饰产物分别在相对分子质量为15×103和31×103处同现条带,提示DBBF对猪血红蛋白的两个亚基进行了分子内共价交联.
