Abstract By using polymerase chain reactionrestriction fragment length polymorphism (PCRRFLP) based Sgene identification system and DNA sequencing technology, the Sgenotypes of ‘Chuxialü’ and ‘Cuiyu’ (Pyrus pyrifolia Nakai. cvs., two of the F1 progenies selected from ‘Xizilü’ × ‘Cuiguan’) were confirmed as S3S4, respectively. The results of fluorescence microscopic observation of pollen germination dynamics in situ and artificial pollination experiment showed that, both ‘Chuxialü’ and ‘Cuiyu’ had middle strength of selfincompatibility, but the crosscompatibility between them were incomplete; the selfincompatibility strength of ‘Cuiguan’ was high, while its crosscompatibility with ‘Chuxialü’ and ‘Cuiyu’ was observed.
