APPLICATION OF DYS Ⅰ DYS Ⅱ,DYS Ⅲ IN LINKAGE AND ALLELE FREQUENCY ANALYSIS OF BMD/DMD GENE

PCR technique was applied to amplification of three polymorphic short tandem repeats(DYS Ⅰ,DYS Ⅱ, DYS Ⅲ) of DMD gene. The PCR products were electrophoresed on polyacrylamide gel. The results showed that the loci were highly polymorphic,with heterozygosities of 76.7%,86.7% and 43.3% in southeast China. Polymorphism Information Contents(PIC) ranged from 0.659 to 0.697. In a large DMD family haplotype linkage analysis demons-traied that there were no recombination in 10 informative meiosis analysed between the three loci and dystrophin gene. In addition,though the loci were highly polymorphic,in some cases the polymorphic loci were homozygotic and could not provide any information for linkage analysis. It indicated that in a linkage locus.several polymorphic markers should be used,so that more accuracy information would be provided.

THE REGULATION OF ENDOGENOUS OPIATE PEPTIDE ON CARDIOVASCULAR EFFECTS OF CAUDAL VENTROLATERAL MEDULLA EXCITATION

The regulative effect of deep peroneal nerve(DPN) inputs on cardiovascular deceleration and depression responses induced by electric stimulation of rabbit's caudal ventrolateral medulla(CVLM) was observed. Experiments were performied on 30 white male rabbits anaesthetized with chloralose-urethane under artifical ventilation. The results showed that:(1)the inputs of DPN could inhibit the cardlac deceleration response evoked by electric stimulation of CVLM(P<0.0001), and the regulative effectsof DPN disappeared in 25~30 min;(2) afteriv atropine(0.25 mg/kg) or scopolamine(0.25 mg/kg) the heart rate and blood pressure reduction value had no significant difference when stimulated CVLM+DPN compared with control group(P>0.05);atropine or scopolamine could not abolish the inhibitory effect of DPN inputs;(3)after tv naloxone(1.5 mg/kg) or microinjection of naloxone into rostral ventrolateral medulla(RVLM)(2μg in 1 μl NS perside bilaterally),the inhibitory effect of DPN inputs was abolished partly(P<0.01).These results suggest that:(1) the cardiac deceleration and depression responses elicited by CVLM excitation in rabbits can be regulated by inputs of DPN;(2)it seems that the regulative function may have no relationship with acetylcholine system. On the other hand, endogenous opiate peptide in RVLM may take part in inhibitory effect of DPN inputs on cardiovascular effects excited by CVLM.

VALUES OF BRAINSTEM AUDITORY EVOKED POTENTIAL IN THE ETIOLOGICAL ANALYSIS AMONG CHILDREN WITH LANGUAGE DEVELOPMENT DISORDER

An evaluation of brainstem auditory evoked potential(BAEP) was carried out in 368 children with language development disorder and in 116 control children matched for age. An abnormal BAEP was documented in 290 children with language development disorder, whereas none was found in control subjects. Among patients with abnormal BAEP, 90 had a history of receiving aminoglycoside antibiotics, 55 had perinatal asphyxia,hypoxic-ischemic encephalopathy or severe infection, 35 had cerebral pulsy, 41 had mental retardation to varied degree, 12 had a history of acute central nervous system infection,and 7 had a family history of hearing loss.Results suggested that BAEP evaluation might be a simple, noninvasive, objective and reliable test. It could be used as a screening test for early detection of hearing loss among children with language development disorder.

CHANGES OF SERUM TOTAL BILE ACIDS IN NEONATAL SEPTICEMIA AND ITS CLINICAL SIGNIFICANCE

Serum concentrations of total bile acids(TBA) in newborns with septicemia and correlation of those concentrations with the results of other routine liver tests including alanine aminotransferase(ALT),total bilirubin(TB), and γ-glutamyl transpeptidase(γ-GT) were studied in 77 cases of neonatal septicemia, with 19 non-infections newborns as controls. The results showed that serum TBA levels significantly increased in septicemia as compared with those of controls(P<0.01) and that serum TBA concentrations in 35 elevated cases sharply declined 7~10 days after effective antibiotic therapy(P<0.01). In addition, we found that TBA concentration in septicemia with septic shock was significantly higher than that with non-septic shock(P<0.05).There were no correlations between the concentration of serum TBA and those of ALT,TB and γ-GT. The study suggested that serum TBA levels in neonatal septicemia remarkedly elevated during the acute stage and apparently decreased during the convalescent stage. Serum TBA levels might reflect the seventy of neonatal septicemia to a certain extent.

COMPARISON OF RECOVERY FOLLOWING PROPOFOL ANESTHESIA WITH RECOVERY FROM ISOFLURANE ANESTHESIA

Propofol is a new intravenous anesthetic. To evaluate its recovery property, 26 patients were divided into two qroups undergoing propofol anesthesia(group P, n=13) and isoflurane anesthesia(group I, n=13). Recovery assessment was based on Post-Anesthesia Care Unit(PACU) scores recorded by nurses. The results showed that the scores of qroup P were remarkably higher in early stage of recovery than those of group I, indicat in that recovery from propofol anesthesia was more rapid and complete.

We have used papaverine and hot water to treat 45 cases of female urethral syndrome by in travesical instillation, as a new and simple method. Among these cases 84.4%(38/4d) were completely relieved or markedly improved. Urodynamic study showed a significant increase of first sensation volume and maximum capacity and other parameters. The symptom and Urodynamic parameters of non-effective cases were improved in some degree as well.
