APPLICATION OF DYS Ⅰ DYS Ⅱ,DYS Ⅲ IN LINKAGE AND ALLELE FREQUENCY ANALYSIS OF BMD/DMD GENE

PCR technique was applied to amplification of three polymorphic short tandem repeats(DYS Ⅰ,DYS Ⅱ, DYS Ⅲ) of DMD gene. The PCR products were electrophoresed on polyacrylamide gel. The results showed that the loci were highly polymorphic,with heterozygosities of 76.7%,86.7% and 43.3% in southeast China. Polymorphism Information Contents(PIC) ranged from 0.659 to 0.697. In a large DMD family haplotype linkage analysis demons-traied that there were no recombination in 10 informative meiosis analysed between the three loci and dystrophin gene. In addition,though the loci were highly polymorphic,in some cases the polymorphic loci were homozygotic and could not provide any information for linkage analysis. It indicated that in a linkage locus.several polymorphic markers should be used,so that more accuracy information would be provided.

VALUES OF BRAINSTEM AUDITORY EVOKED POTENTIAL IN THE ETIOLOGICAL ANALYSIS AMONG CHILDREN WITH LANGUAGE DEVELOPMENT DISORDER

An evaluation of brainstem auditory evoked potential(BAEP) was carried out in 368 children with language development disorder and in 116 control children matched for age. An abnormal BAEP was documented in 290 children with language development disorder, whereas none was found in control subjects. Among patients with abnormal BAEP, 90 had a history of receiving aminoglycoside antibiotics, 55 had perinatal asphyxia,hypoxic-ischemic encephalopathy or severe infection, 35 had cerebral pulsy, 41 had mental retardation to varied degree, 12 had a history of acute central nervous system infection,and 7 had a family history of hearing loss.Results suggested that BAEP evaluation might be a simple, noninvasive, objective and reliable test. It could be used as a screening test for early detection of hearing loss among children with language development disorder.
