内阿片肽对延髓尾端腹外侧心血管中枢活动的调节

摘要:

用弱电流刺激延髓尾端腹外侧区(CVLM)造成心率减慢、血压下降模型,观察腓深神经(DPN)传入冲动对CVLM兴奋引起心血管效应的调节作用,并分析其机制。实验在30只脲酯(500mg/kg)加氯醛糖(50mg/kg)混合静脉麻醉、三碘季铵酚制动、人工呼吸的家兔上进行。实验发现:①DPN的传入冲动对CVLM兴奋所引起的心率减慢有抑制作用(P<0.0001);②静脉注射阿托品或东莨营碱(0.25mg/kg)后,刺激DPN合并刺激CVLM时,心率及血压下降值与对照组相比无显著差异(P>0.05),不能阻断DPN传入冲动对CVLM兴奋效应的调节;③静脉注射纳洛酮(1.5mg/kg),以及延髓头端腹外侧区(RVLM)微量注射纳洛酮两侧各2μg(1μl),均可阻断DPN传入冲动对CVLM的调节(P<0.01)。实验结果提示:DPN的传入冲动可对CVLM兴奋时的心血管效应进行调节,此调节作用似与乙酰胆碱系统无关,而很可能是由RVLM部位的内阿片肽参与作用所致。

Abstract:

The regulative effect of deep peroneal nerve(DPN) inputs on cardiovascular deceleration and depression responses induced by electric stimulation of rabbit's caudal ventrolateral medulla(CVLM) was observed. Experiments were performied on 30 white male rabbits anaesthetized with chloralose-urethane under artifical ventilation. The results showed that:(1)the inputs of DPN could inhibit the cardlac deceleration response evoked by electric stimulation of CVLM(P<0.0001), and the regulative effectsof DPN disappeared in 25~30 min;(2) afteriv atropine(0.25 mg/kg) or scopolamine(0.25 mg/kg) the heart rate and blood pressure reduction value had no significant difference when stimulated CVLM+DPN compared with control group(P>0.05);atropine or scopolamine could not abolish the inhibitory effect of DPN inputs;(3)after tv naloxone(1.5 mg/kg) or microinjection of naloxone into rostral ventrolateral medulla(RVLM)(2μg in 1 μl NS perside bilaterally),the inhibitory effect of DPN inputs was abolished partly(P<0.01).These results suggest that:(1) the cardiac deceleration and depression responses elicited by CVLM excitation in rabbits can be regulated by inputs of DPN;(2)it seems that the regulative function may have no relationship with acetylcholine system. On the other hand, endogenous opiate peptide in RVLM may take part in inhibitory effect of DPN inputs on cardiovascular effects excited by CVLM.
