作者利用抗Hb-HRP建立免疫斑点法(Dot-ELISA)粪便潜血试验,检测了46例正常人和35例大肠癌患者的粪便标本,并与RPHA粪潜血试验相对照.结果发现,Dot-ELISA检测人Hb的灵敏度为1 μg/ml,而50 μg/ml的羊、鸡、鸭、免、鼠Hb用本法检测均为阴性.46份正常人粪便标本的阳性率为6.5%(3/46);35份大肠癌患者粪标本的阳性率为88.6%(31/35).与RPHA法对比表明,Dot-ELISA用于检测粪潜血以筛检大肠癌,与RPHA法同样具有较好的敏感性和特异性.
Using anti-Hb antibody-HRP conjugation, the authors have developed a Dot-ELISA method for detection of fecal occult blood. The result shows that the lower detection limit of Dot-ELISA for human Hb was 1 μg/ml, while that for the Hb of chickens, sheep, ducks, rabbits and mice (went up to a level of 50 μg/ml) with negative results. The positive rate of Dot-ELISA FOB test for stool samples collected from 46 healthy adults and 35 colorectal cancer patients were 6.5% and 88.6%, respectively.Comparing with the RPHA FOB test, the Dot-ELISA FOB test shows a similar sensitivity and specifity in detection of occult blood and possesses the advantages of rapidity, simplicity, low cost, objectiveness and accuracy, and so it is considered to be more suitable for the large-scale screening program of colorectal cancer.
