摘要:

从蝮蛇毒中提取磷酸二酯酶,用此酶免疫BALB/c小鼠,取其脾细胞与小鼠骨髓瘤细胞Fo融合,以间接ELISA检测杂交瘤细胞培养上清液和小鼠腹水中的特异性抗体,其效价分别为1:128和1:51200.抗原阻断试验结果表明,此抗体对蛇毒磷酸二酯酶具有特异性。该杂交瘤细胞株定为G8,该株单抗属鼠IgG2a亚型,经体外持续培养6个月,其分泌抗体性能稳定。

Abstract:

Phosphodiesterase extracted from the venom of Agkistrodon blomhoffii brevicaudus was used to immune BALB/c mice,and their splenic cells were fused with mouse myeloma cell Fo. Ahybridoma cell strain G8,which could secrete anti-venom-phosphodiesterase antibodies was isolated. The specific antibody in the supernatant of the hybridoma cell cultures and in the ascites of mice innoculated with the hybridoma cells were detected through indirect ELISA,and the titers were 1:128,1:51200 respectively. The results of antigen block test showed that this antibody was specific to the phosphodiesterase of venom. This strain of monoclonal antibody belongs to the mouse IgG2a subtype. Continuous culturing for six months in vitro has revealed the antibody secretion of the cell strain to be stable.
