抗淋球菌单克隆抗体的制备及其生物学特性研究

摘要:

目的:研制抗淋球菌单克隆抗体,为建立新的淋病诊断方法奠定基础。方法:以淋球菌29106-5株作抗原,按常规淋巴细胞杂交瘤技术制备单克隆抗体。并采用ELISA双抗体夹心法研究单抗的特异性和敏感性。结果:建立了5个持续分泌抗淋球菌单抗的杂交瘤细胞株。他们所分泌的抗体效价可达1:8192~1:32786。5株单抗均能与淋球菌发生特异性结合,但对脑膜炎球菌、葡萄球菌、大肠杆菌等15种其他微生物不起反应。淋球菌的最小检测量为1×104个/ml。结论:本研究建立的单抗在检测淋球菌时有高度特异性和敏感性。

Abstract:

Objective:To develop monoclonal antibodies against Neisseria gonorrhoeae for establishing a new diagnostic method of gonorrhoea.Methods:Monoclonal antibodies against strain 29106-5 of N.gonorrhoeae were prepared by the lymphocyte hybridoma technique.The specificity and sensitivity of McAbs were studied using the sandwich method of ELISA with double antibodies.Results:Five cell lines of hybridoma secreting constantly McAbs to N.gonorrhoeae were obtained.The titer of McAbs reached to 1:8192~1:32786.All McAbs could react to N.gonorrhoeae,but failed to react to other microbe such as N.Meningitidis,staphylococcus and E.Coli.The minimum dose of N.gonorrhoeae which can be detected by ELISA was 1×104 bacteria per ml.Conclusion:The sandwich method of ELISA using McAbs to detect N.gonorrhoeae possessed higher specificity and sensitivity.
