基于合作水解模型的微管动态不稳定性研究

摘要: 微管是细胞中重要的组成部分和功能组件,在细胞参与的一系列生命活动中起着极其重要的作用.微管在细胞内外均呈现独特的动态不稳定性.目前关于微管内蛋白之间纵向相互作用对微管动态不稳定性影响的研究逐渐增多.应用合作水解模型得到了微管内蛋白之间纵向相互作用与微管稳定性的关系.结果表明,微管链内纵向相互作用越强,微管末端存在连续GTP微管蛋白片段的概率越小.游离GTP微管蛋白浓度越大,微管链内纵向相互作用对微管末端的影响越强.当游离GTP微管蛋白浓度较低、微管链内纵向相互作用较弱时,微管帽体较长,微管不容易发生塌缩,进而稳定性也较强.

Abstract: Microtubules consist of cytoskeleton multifilament proteins and play significant roles in many fundamental biological processes including cell division, cellular transport and motility. Both inside and outside of the cell, microtubules manifest an unique dynamic instability. In this paper, we investigate the dynamic instability of microtubules using a cooperative hydrolysis model. For this model, any GIPtubulin hydrolyzes with a rate depending on the states of its neighbors, we use α to describe the influences of its neighbors. Using a new theoretical method, we calculate the steady state of its neighbors. Using a new theoretical method, we calculate the steady state of this model. The influences of α to the dynamics instability of microtubules are discussed. The results show that α influences the distribution of Tsubunit clearly, and α influences the cap size remarkably, especially when U is small.
